Hello! I am working with data from published articles and they didn't state the primers that are used (and the co-authors dont reply me back). Some of them merged the sequences with primers still on. I can understand that primers are there because there are repeating sequences in the forward and the reverse, but i cannot find the actual primers in any way. What should I do? Thanks. (V3-V4 datasets)
Hi @Acelya_Dalgic,
Unfortunately I am not familiar with any way to find and remove primers without knowing what they are.
If you can figure out how long the primers are you could use the --p-trim parameter in dada2 to trim them from the beginning of the sequences.
Hope this helps!
