thanks for your positive response and help through out.
It was really easy and just a matter of few minutes to create rooted-tree from unrooted phylogeny.
however overcoming issue other rise
Plugin error from diversity:

None of the sample identifiers match between the metadata and the coordinates. Verify that you are using metadata and coordinates corresponding to the same dataset.

Debug info has been saved to /tmp/qiime2-q2cli-err-ue6k5xgf.log
I think the samples in metadata do not match to the ones in in root file.
How to check the names of samples in these files.

I suspect you're on the right track!

The sample ids in your metadata should be the left-most column in your study metadata file. You can find the sample IDs associated with your data in the visualization produced by qiime demux summarize
