Sequence quality control - Demultiplexed sequence counts summary

Hi, I am trying to follow the Moving pictures tutorial and get the paired-end-demux.qzv.
However, the results are not as imaging.
I saw the figure in the tutorial, the number varies too much, and I don't know how to interpret the difference.

And How to choose the trunc and trim size?
Thank you so much.

So you are downloading and processing the moving pictures data but get a different result? Can you please attach the QZV file?

See the "interactive quality plot" tab in that QZV. Trim/truncate where quality begins to drop off — the moving pictures tutorial and other tutorials, as well as this forum, have much more information on how to decide on where to cut.

I hope that helps!

Hi, @Nicholas_Bokulich,
Thank you.
The figure one is data of mine. Human fecal samples 16S sequence data.
I analysis own data based on the steps of Moving pictures tutorial.
The qzv data atteched before:
paired-end-demux.qzv (287.7 KB)
The sequence counts is very huge, Could you help me to illustrate the difference?
Thank you.
Qiong

You have many sequences, which is a good thing. The tutorial has very few sequences because a small selection was sampled to make a tutorial dataset that would run quickly. So there is no inconsistency here.

Does that answer your question?

Hi, @Nicholas_Bokulich, Thank you for your reply.
You did answer my question, Thank you.
